Formulation & Evaluation Of Transferosome Mefenamic Acid Based Gel For Topical Drug Delivery In Rheumatoid Arthritis

ABSTRACT

Background: Design and characterization of a nanocarrier-loaded topical gel to enhance the localized delivery and therapeutic efficacy of mefenamic acid (MA) for rheumatoid arthritis (RA). Nanostructured delivery platforms advance drug transport by increasing absorption and efficacy, whilst topical gels provide focused relief with fewer adverse effects than oral drugs. Aim: This research aimed to design, formulate, conduct, and evaluate studies on a nanocarrier-loaded gel for optimizing the topical penetration and therapeutic availability of mefenamic acid (MA) in rheumatoid arthritis (RA) management. Methods: Mefenamic acid-loaded nanoparticles were formulated by combining the polymers. Utilizing FTIR and DSC techniques, a drug and polymer compatibility evaluation was carried out. The nanoparticles were analyzed to determine their size, surface structure, capacity to encapsulate drugs, drug release, release rate, and stability. Results: The composition and performance of transferosomes with Mefenamic acid (100 mg per batch). showed the Transferosomes shows the highest drug release (98.4%) and entrapment efficiency (91.04±0.24%), despite having the largest particle size (8.96±0.45μm). The results highlight the influence of Carbopol, Tween, and ethyl cellulose content on vesicular systems' size, entrapment and release properties. Conclusion: Transferosomes loaded gel with Mefenamic acid (100 mg) were evaluated for particle size, entrapment efficiency, and drug release. Transferosomes showed superior drug release and efficacy, highlighting their potential in managing rheumatoid arthritis via topical delivery..
